Rao, G. Ramananda (Indian Institute of Science, Bangalore, India), M. Sirsi, and T. Ramakrishnan. Enzymes in Candida albicans. II. Tricarboxylic acid cycle and related enzymes. J. Bacteriol. 84:778-783. 1962.-Evidence is presented to show the operation of the tricarboxylic acid cycle in Candida albicans, by studies with whole cells, cell-free preparations, and by the demonstration of most of the enzymes involved in the cycle. Cell-free extracts contained the following enzymes: condensing enzyme; aconitase; isocitric, alpha-ketoglutaric, succinic, and malic dehydrogenases; malic enzyme; fumarase; reduced diphosphopyridine nucleotide (DPNH) oxidase; DPNH-cytochrome c reductase; reduced triphosphopyridine nucleotide (TPNH) cytochrome c reductase; and diaphorase. Pyruvic dehydrogenase, TPNH oxidase, and transhydrogenase activities could not be detected under the test conditions.

Candida albicans transcription factor Ace2 regulates metabolism and is required for filamentation in hypoxic conditions.
Ace2 transcription factor family genes are found in many fungal genomes and are required for regulation of expression of genes involved in cell separation. We used transcriptional profiling to identify the targets of Ace2 in Candida albicans, and we show that these include several cell wall components, such as glucanases and glycosylphosphatidylinositol-anchored proteins. Expression is downregu...
متن کاملMetabolomic profiling for the identification of potential biomarkers involved in a laboratory azole resistance in Candida albicans
Candida albicans, one of the most common fungal pathogens, is responsible for several yeast infections in human hosts, being resistant to classically used antifungal drugs, such as azole drugs. Multifactorial and multistep alterations are involved in the azole resistance in Candida albicans. In this study, a FCZ-resistant C. albicans strain was obtained by serial cultures of a FCZ-susceptible C...

متن کاملInhibitors of the Glyoxylate Cycle Enzyme ICL1 in Candida albicans for Potential Use as Antifungal Agents
Candida albicans is an opportunistic pathogen that causes candidiasis in humans. In recent years, metabolic pathways in C. albicans have been explored as potential antifungal targets to treat candidiasis. The glyoxylate cycle, which enables C. albicans to survive in nutrient-limited host niches and its. Key enzymes (e.g., isocitrate lyase (ICL1), are particularly attractive antifungal targets f...
متن کاملReversible transdominant inhibition of a metabolic pathway. In vivo evidence of interaction between two sequential tricarboxylic acid cycle enzymes in yeast.
The enzymes of the Krebs tricarboxylic acid cycle in mitochondria are proposed to form a supramolecular complex, in which there is channeling of intermediates between enzyme active sites. While interactions have been demonstrated in vitro between most of the sequential tricarboxylic acid cycle enzymes, no direct evidence has been obtained in vivo for such interactions. We have isolated, in the ...
